Herbal medicines are now commonly used in illness treatment. These herbs and modern medicines can cause the herbal-drug interaction by affecting the Cytochrome P450 enzyme function. This research emphasized on the effect of herbal medicines onto rat CYP3A4 enzyme, a member of Cytochrome P450. Herbal-crudes were extracted in Deionized water. High Performance Liquid Chromatography (HPLC) technique was used to determine the amount of 6-ß hydroxy testosterone, major metabolite of 6-ß testosterone hydroxylation reaction by CYP3A4 enzyme. Then convert into percentage of inhibition. The R2 of 6-ß hydroxy testosterone standard curve used was 0.9936. The results revealed that the aqueous extract of 3 herbs, Lawsonia inermis (Tien Dange), Nymphaea lotus L. var pubescens Hook.f. & Th. (Boa Khom) and Punica granatum (Tub Tim) has no inhibition activity whereas inhibition degree of Cyperus rotundus (Hawe Moo) and Allium ascalonicum (Hom Dange) are found to be 42.15 and 12.24 % respectively.
