The investigation of effects on growth and accumulation of canthin-6-ones such as 9-hydroxycanthin-6-one and 9-methoxycanthin-6-one in root cultures of EL were studied. Effect of culture media supplemented with 3%(w/v) sucrose such as MS, WP and B5 showed that root cultures produced the highest biomass in half strength of B5 basal medium (0.29 g DW). Accumulation of 9-hydroxycanthin-6-one and 9-hydroxycanthin-6-one were found in the highest on B5 medium (1.440.03 mg/g and 1.420.03 mg/g, respectively). Effect of carbon sources in ½WP supplemented with 3% of different carbon sources such as sucrose, glucose and fructose showed that root cultures produced the highest biomass and yield of 9-hydroxycanthin-6-one at 60 days of culture time in 3% sucrose (0.29 g DW, 0.810.02 mg/g) and fructose (0.30 g DW, 0.760.03 mg/g). The content of 9-hydroxycanthin-6-one was highest in 3% Fructose (1.440.03 mg/g) at 40 days of culture time and Sucrose (0.930.01 mg/g) at 20 days of culture time. In addition to effect of elicitors such as methyl jasmonate, chitosan, sodium biphosphate (Na2HPO4) and yeast extracts showed that the accumulation of canthin-6-one tends to increase at 6 days after elicited. The content of 9-hydroxycanthin-6-one was highest in 5mg/L Na2HPO4 (2.360.01mg/g) elicitor by 0.98-fold and 9-methoxycanthin-6-one was highest in 0.5mg/L yeast extracts (1.050.001 mg/g) elicitor by 2.23-fold.
