The simultaneous analysis of 7 biogenic amines (cadaverine, putrescine, histamine, spermine, spermidine, tryptamine and tyramine) as their 9-Fluorenymethyl chloroformate (FMOC) derivatives prior to reversed phase high performance liquid chromatography with gradient elution of 0.1% phosphoric acid in methanol and water and fluorescence detection has been studied. Parameters affect the analysis such as derivatization procedure and gradient elution profile were investigated. The derivatization procedure was unsuitable due to it produced various derivativescause many interference peaks occur on chromatogram. The optimum gradient elution profiles was developed to solve this problem, all of major peaks were separated by this profile. However, the proposed conditions were inappropriate to be used in the analysis of real samples. The method should be developed for further analysis. |
