Inflammation is a response of immunological system to injury or exogenous substances leading to diseases that affect quality of life such as gout or rheumatoid arthritis. Several studies reveal the anti-inflammatory effect in various types of mushroom. In this study, we aim to determine the anti-inflammatory effect on consumable mushrooms Objectives= to evaluate the anti-inflammatory effect of L. edodes, P. ostreatus and P.pulmonarius extracts on white blood cell (U937 cell) Material and Method= The cytotoxicity in water extract of three mushrooms at the concentration of 1.87, 3.75, 7.5, 15, 30, 60 and 120 µg/mL and CHCl3= CH3OH extract at the concentration of 30, 60,120 and 240 µg/mL were determined by MTT assay and the anti-inflammatory effect of the mushroom extracts were investigated by Real-Time PCR. To evaluate the mechanism of anti-inflammation, the expression of TNF-α, IRF5 and TGF-β were used as the represented genes for inflammation, macrophage polarization and inflammation suppression respectively. Result= Cytotoxicity test showed that the treatment of water extracts of P. pulmonarius and P. ostreatus at concentration of 120 µg/mL in 48 hours significantly decreased amount of cells (P<0.05). In 72 hours, treatment of P. pulmonarius and P. ostreatus extracts at concentration >60 and >7.5 µg/mL significantly decreased amount of cells, respectively (P<0.05). There was no significantly decrease amount of cells at concentration ≤120 µg/mL in water extract of L. edodes. CHCl3= CH3OH extract of L. edodes at concentration 240 µg/mL significantly decreased amount of cells in 48 hours (P<0.05) while the treatments of P. pulmonarius and P. ostreatus were not found cytotoxic activity. The anti-inflammatory effect at 30 µg/mL water extract showed in the expression ratio of TNF-α and IRF5 and TGF-β when compared to non-treatment group. After 48 hours, the gene expression ratio of TNF-α and IRF5 and TGF-β were 0.79, 3.18 and 7.97-fold in L. edodes treatment, 3.06, 12.77 and 2.29-fold in P. pulmonarius treatment and 0.53, 82.71 and 17.27-fold in P. ostreatus respectively. Conclusion= the water extracts of P. pulmonarius and P. ostreatus at concentration of 120 µg/mL significantly decreased amount of cells in 48 hours, P. pulmonarius and P. ostreatus extracts at concentration of >60 and >7.5 µg/mL significantly decreased amount of cell in 72 hours, respectively. There was no significantly in L. edodes treatment. We found no significantly decrease amount of cells in CHCl3=CH3OH extract except 240 µg/mL L. edodes extract in 48 hours. All extracts had no effect on macrophage polarization, while P. ostreatus exhibited the most anti-inflammatory effect considering by decrease of TNF-α and increase of TGF-β.
