| Neuron cells and nervous system are important for the human. When they were destroyed by various factors Such as free radicals, chemicals lead to Neurodegenerative disease. Finding to protect the neuron cells are necessary to prevent Neurodegenerative disease. Objective= The study was aimed to study neuroprotective effect of Careya sphaerica Roxb.extracts in hydrogen peroxide (H2O2) induced SHSY5Y neuroblastoma cells. Methods= Careya sphaerica Roxb.extracts included methanol extract of leave and flower and butanol extract of flower to investigate antioxidant activity by DPPH assay and ABTS assay. To study neuroprotective effect were determined by WST-8 assay.To investigate effect of Careya sphaerica Roxb.extract on Caspase-3 that is marker of apoptosis by western blot. Result= Antioxidant activity by DPPH assay and ABTS assay. The results showed that extracts possess antioxidant activity, IC50 of Buthanol extract of flower and methanol extract of flower and leaves were 40.25 ± 8.10, 43.82 ± 11.48, 41.58 ± 2.37 and 45.94 ± 8.78, 42.41 ± 10.94, 41.55 ± 10.38 respectively. The results of neuroprotective effects that determined by WST-8 assay were buthanol extract of flower possess the most potent effect. Effect of Careya sphaerica Roxb.extracts on caspase-3 activation in H2O2 induced-neuronal cell death by Western blot analysis. The results showed that H2O2 induced caspase-3 activation by cleaved caspase-3 appearance. The extracts protected H2O2-induced neuronal cell death by caspase-3 inhibition. Conclusion= Careya sphaerica Roxb.Extracts including methanol extract of leave and flower and butanol extract of flower reveals neuroprotective effects on H2O2-induced neuron cell. |
