Title : EFFECT OFVERNONIA CINEREA AND 5 THAI HERBAL CRUDE EXTRACTS ON RAT CYP3A4 ENZYME ACTIVITY IN-VITRO

This research aimed to study the effect of 6 Thai herbal crude extracts on function of rat CYP3A4 enzyme. Metabolic test were carried out in vitro using testosterone as a specific CYP3A4 probe substrate. The amount of 6beta-hydroxy-testosterone obtained from metabolic reaction with or without herbal crude extract was determined using HPLC then converted into %inhibition. Result revealed that the aqueous extract of 6 herbs inhibit CYP3A4 enzyme i.e. Pi-kun (MimusopselengiL.) 46.68%, Ngiew (Bombaxanceps Pierre.) 51.32%, Pai(Bambusabambos) 61.83%, Ton-aor (Arundodonax) 77.33%, Nard (Blumeabalsamifera) 58.55% andYa-dok-khao(Vernoniacinerea) 89.23%. The methanol extracts of 5herbs inhibitCYP3A4 activity i.e.Pi-kun 65.51%, Ngiew71.96%, Pai 76.42%, Nard71.21% and Ya-dok-khao80.25%. The hexane extract of 6herbs also inhibit CYP3A4 activity i.e.Pi-kun 53.98%, Ngiew55.94%, Pai58.83%, Ton-aor85.72%, Nard13.77%and Ya-dok-khao 62.74%. It could be conclude that all 6 Thai herbs inhibit CPY3A4 function in different degree.
