Title : THE EFFECT OF POUTERIA CAMPECHIANA FRUIT EXTRACTS TO INHIBIT THE GROWTH OF CANCER CELLS.

Cancer is harmful disease many anti-cancer drugs came from plants which have efficacy and safety. Objective= To test the effect of pouteria campechiana fruit extracts to inhibit the growth of cancer cells and to investigate molecular mechanism by Pouteria campechiana fruit extracts induced apoptosis. Materials and methods= MTT assay were studied % cell viability of HT-29 cell by Pouteria campechiana fruit extracts compare with doxorubicin (positive control) and without Pouteria campechiana fruit extracts (negative control) and % cell viability of FHC by Pouteria campechiana fruit extracts compare with doxorubicin (positive control) and without Pouteria campechiana fruit extracts (negative control).MTT assay was determined by using UV spectrophometer technique then converted into % cell viability. Moreover morphologic studied of HT-29 and FHC cells by Phase contrast microscopy The mechanism of pouteria campechiana fruit extracts to inhibit the growth of cancer cells studied by Western blot Result= MTT assay= revealed that at 48 hours Pouteria campechiana fruit extracts concentration 100 µg/ml % cell viability of HT-29 cell was 41% (P<0.01) and 72 hours Pouteria campechiana fruit extracts concentration 10 µg/ml, 100 µg/ml % cell viability of HT-29 cell was 40%(p<0.01) and 41%(P<0.01). % cell viability of FHC cells treated with Pouteria campechiana fruit extracts at 24, 48 and 72 hours were more than 90% and not decrease when higher concentration. Phase contrast microscopy= HT-29 cells were shrinkage and fragmentation these were apoptotic morphology. Western blot= founded caspase-3 (the marker of apoptosis). Conclusion= Pouteria campechiana fruit extract inhibited HT-29 cell proliferation by apoptosis through apoptosis inducing proteins.
