A key characteristic of cancer is the rapid creation, leading to uncontrolled cell growth; apoptosis is inhibited. Cancer cells havetheability to migrate and invade the surrounding tissue, resulting in cancer cell metastasis to other organs or other part of body. Thus, inhibition of proliferation, migration and invasion of cancer cells are the important goalsof the treatment of cancer. Objectives: 1) To study the inhibitory activity on proliferation of colon cancer cell (HT-29), and liver cancer cell (Hep-G2), were compared with colon normal cell (FHC). 2) To study the inhibitory activity on cell migration and cell invasion of colon cancer cell. Methods: 7-methoxyheptaphylline (7-MH) was an active constituent derived from “Song-fa” (Clausena harmandiana)which was evaluated for cell cytotoxicity at various concentration and time by using MTT assay, and Doxorubicin was used as a standard compound. The effects of 7-MH on cell migration and cell invasion were investigated by using Trans well migration assay and Trans well invasion assay respectively. In addition, the pathways of the proliferation, the migration and the invasion of 7-MH were investigated to find out the signaling protein by means of Western blot analysis.Results:7-MH 100 µM showed the cytotoxicity toward HT-29 and Hep-G2, which was significantly different from the control group (p<0.01) by % inhibition=53.38 ± 10.48, 73.85 ± 10.82µM (HT-29) and 31.83 ± 2.43, 48.69 ± 4.20µM (HepG2) at 24 and 48 hours respectively, while no cytotoxic effects on FHC was found. Moreover, 7-MH showed the inhibitory activity on migration and invasion of HT-29 in concentration and time dependent manner. 7-MHwas inhibited HT-29 proliferation by inhibiting anti-apoptotic protein (Bcl-2, Bcl-xL, survivin) through MAPK/Erk pathway (Erk1/2) and were inhibitedmigration and invasion by Matrix metalloproteinase-9 (MMP-9) inhibition.Conclusion: 7-MH demonstrated the inhibitory activity on proliferation of colon cancer cell and showed inhibitory activity on migration and invasion of colon cancer cell in relation to metastasis of cancer.
