Now many herbal productsare very popular. Some herbal products could be metabolizedspecifically by CYP3A4 enzyme.Thus using medications and herbal products together may cause of drug interaction.Objectives:This research aimed to study the effect of 3 Thai herbal crude extracts on function of rat CYP3A4 enzyme. Methods:Metabolic test were carried out in vitro using testosterone as a specific CYP3A4 probe substrate. The amount of 6beta-hydroxy-testosterone obtained from metabolic reaction with or without herbal crude extract was determined using HPLC then converted into %inhibition. Result: We found that the aqueous extract of 3 herbs also inhibit CYP3A4 activity i.e. Ma-li (Jasminum Sambac (L.) Aiton) 37.80%,Bua-luang(Nelumbo nucifera Gaertn.)32.00% and Sa-ra-pee (Mammea siamensis (Miq.) T.)25.70%.Themethanol extracts of 3 herbs also inhibit CYP3A4 activity i.e.Ma-li (Jasminum Sambac (L.) Aiton) 21.91%,Bua-luang(Nelumbo nucifera Gaertn.) 15.67% and Sa-ra-pee (Mammea siamensis (Miq.) T.)62.39% The hexane extract of 3 herbs also inhibit CYP3A4activity i.e.Ma-li (Jasminum Sambac (L.) Aiton) 46.88%, Bua-luang (Nelumbo nucifera Gaertn.) 66.15% and Sa-ra-pee (Mammea siamensis (Miq.) T.) 63.90%.Conclusions:It could be conclude that 3 Thai herbs inhibit CPY3A4 function in different degree. |
