The objective of this study was to develop a Theophylline effervescent dry powder for pulmonary drug delivery. The techniques used to reduce the particle size in order to increase solubility were spray-drying and ball mill. Theophylline effervescent dry powder formulas from Phase 1 were utilized as master formula and reproduced in this study. Spray-drying was run with inlet air 200 °C, outlet air 90 °C and flow rate 2 mL/min. While ball mill was run of using the diameter of 30 mm (200 rpm for 50 minutes). Physical characteristics of Theophylline effervescent dry powder from 2 different techniques were then compared under a microscope. Polymorphism change was studied using Differential Scanning Calorimetry. Theophylline concentration after effervescent reaction occurred was determined by HPLC. The results indicated that particles prepared by spray-drying were smaller than those of ball mill. More over spray-drying technique produced more spherical particles than ball mill and gave faster dissolving rate in water. Theophylline concentration was determined by HPLC found unchanged after the effervescent reaction. Future study should focus on preparation method of effervescent dry powder, study of an appropriate dose, pharmacokinetic and drug delivery efficiency.
