White Kwao Krua (Pueraria candollei var. mirifica Airy Shaw et. Suvat or Pueraria mirifica ) is a local herb of Thailand. In Thai traditional medical, the tuberous root used for healthy supplement. The tuber of this species contain phytoestrogen subsatance included miroestrol, deoxymiroestrol, daidzin, daidzein, genistein, genistin. This study interested in isolation of miroestrol and deoxymiroestrol. The method for purification of miroestrol and deoxymiroestrol was patented and complicated process. The objective of this study aim to develop the comfortable method for purification of miroestrol and deoxymiroestrol. 5.15 kg dried tuber cortex of Pueraria mirifica was macerated with hexane, ethyl acetate, and methanol, respectively .Then ethyl acetate extract was isolated by Column chromatography technique using silica gel 60 and hexane=ethyl acetate as a mobile phase. The fraction containing miroestrol and deoxymiroestrol were further separated by Gel filtration chromatography using sephadex LH 20 with the mobile phase as ethyl acetete and methanol. This method can purify miroestrol and deoxymiroestrol easily than previously reported.
