Herbal medicines are widely used for treatment of common illnesses either as a single drug or combination with other modern drugs. It is possible to cause herbal-drug interaction by affecting to cytochrome P450 enzyme function. This research aimed to study the effect of Kot-Chulalampha (Artemisia pallens) and other 4 Thai herbal crude extracts on the function of rat CYP3A4 enzyme. Metabolic tests were carried out in triplicate in vitro using Testosteron (TST) as a specific CYP3A4 probe substrate. The amount of 6β-hydroxy testosterone (6β-TST), major metabolite of TST, obtained from each metabolic reaction with or without herbal crude extracted was determined using High Performance Liquid Chromatography (HPLC) technique, then converted into %inhibition. Results revealed that the aqueous extracts of 3 herbs were affected CYP3A4 enzyme function in different degree of inhibition i.e. Kot-pungpla (Dischidia rafflesiana) 25.76%, Kot-Chulalampha (Artemisia pallens) 11.92%, and Kot-Huabua (Ligusticum chuanxiong) 11.48%. The alcoholic extracts of 2 herbs strongly inhibited CYP3A4 (i.e. Kot-pungpla 91.03% and Kot-Chulalampha 76.90%) meanwhile alcoholic extract of Kot-Huabua has mild inhibitory effect (6.03%). The hexane extracts of 4 herbs also inhibited CYP3A4 activity (i.e. Kot-Huabua 41.96%, Kot-pungpla 30.22%, Kot-Chulalampha 25.25%, and Tienkedhoy 5.91%) whereas the extract of Kot-kema has no inhibitory effect. It could be concluded that 4 Thai herbs inhibit in vitro CYP3A4 function in different degree. In addition, Kot-kema extracts do not inhibit CYP3A4 at the curtain condition test at all.
