The aim of this study was develop of probiotics entrapment in microspheres and solve a problem survival of probiotics in gastrointestinal tract. This study used Lactobacillus casei ATCC 334 entrapped in calcium alginate bead by droplet extrusion/orifice method. Then device a part of alginate bead to coated with gelatin (1mg/ml) 4 hours. All bead were use in this study for evaluation properties of microspheres. The results showed that gelatin-alginate beads had % viability 25% compare with uncoated gelatin-alginate beads. Both type of beads not swelled in pH 1.2 but uncoated gelatin-alginate beads swell and burst (60 minutes) slower than gelatin-alginate beads (30 minutes) in pH 7.4. In conclusions, Both type of bead was able to release in intestinal tract but not release in stomach. There for, probiotics have conducted to diliver and targeted release in gastrointestinal tract.
